Internal Reference:
KM-XCS-1
Undefined basement membrane extracts make every new lot a small experiment. Xbind replaces that uncertainty with a precisely defined surface for human iPSCs in feeder-free culture. Consistent composition means consistent performance from lot to lot, without pre-screening, and without the risk of introducing adventitious agents or xenogeneic antigens.
The active component is a high-purity synthetic ECM peptide engineered for complete integrin-binding activity. It engages iPSC surface integrins, including α6β1 and αvβ3/αvβ5, activating signaling that supports survival, adhesion, and self-renewal. Cells typically attach within 30 to 60 minutes. iPSCs cultured on Xbind remain positive for OCT4, NANOG, SSEA-4, and TRA-1-60, maintain a normal karyotype, and retain trilineage differentiation potential over multiple passages.
Recommended use: Feeder-free attachment, expansion, and long-term maintenance of human iPSCs.
Applications
- Feeder-free iPSC attachment
- iPSC expansion
- Long-term iPSC maintenance
- Flexible coating: 1 hour, 3 hours, or overnight

Recommended coating volumes
| Vessel | Growth area | Volume |
|---|---|---|
| 96-well plate | 0.32 cm² | 50 to 75 µL per well |
| 24-well plate | 1.9 cm² | 200 to 250 µL per well |
| 12-well plate | 3.8 cm² | 400 to 500 µL per well |

| T-75 flask | 75 cm² | 7.5 to 9.0 mL |
Coating steps
- 1Mix. Invert the vial gently until homogeneous. Do not vortex.
- 2Add. Add the recommended volume and rock gently so the whole growth surface is covered.
- 3Incubate. Choose one: 37°C for 1 hour, room temperature (15°C to 25°C) for 3 hours, or 4°C overnight (12 to 18 hours).
- 4Aspirate. Remove the residual coating solution completely. Do not rinse.
- 5Seed. Immediately seed iPSCs in your preferred medium at the desired density.
- 6Distribute. Rock gently to spread cells evenly and place in a humidified incubator at 37°C, 5% CO₂.
Quality Control
Every lot is tested before release:
- Purity: active component > 95% pure by SDS-PAGE
- Endotoxin: ≤ 750 EU/mg
- Sterility: pass (USP <71> / EP 2.6.1)
- Mycoplasma: negative by qPCR / culture-based assay
- Functional activity: pass, iPSC adhesion and proliferation bioassay
Lot-specific results are on the Certificate of Analysis. Email us to request the CoA and include your lot number from the bottle or vial label.
Storage & Shipping
| Shipping Condition | Gel ice packs |
| Storage Temperature | 2°C to 8°C, upright. Protect from light; keep in the original container. |
| Stability | 6 months from the date of manufacture, unopened, when stored correctly. Expiry date is on the vial label. |

Frequently Asked Questions
Do I need to dilute Xbind?
No. It is supplied ready to use. Do not dilute.
Should I rinse the plate after coating?
No. Aspirate the coating solution completely and seed immediately. Rinsing reduces surface activity.
Which coating time should I use?
Any of the three: 37°C for 1 hour, room temperature for 3 hours, or 4°C overnight (12 to 18 hours). Pick the one that fits your schedule.
Do I need to pre-screen new lots?
Xbind has a defined composition, so lot-to-lot pre-screening is not needed the way it is with undefined basement membrane extracts. Concentration and performance data for each lot are on the Certificate of Analysis.
My vial arrived frozen.
Do not use it. Freeze-thaw degrades the active component. Contact us for a replacement.
