Internal Reference:
KM-UDS-1
Trypsin gets the job done, but it is hard on cells. It strips surface proteins, and a few extra minutes in the incubator can cost you viability. Universal Detachment Solution takes a gentler route. Its mild enzymatic action releases adherent cells from the vessel while helping preserve cell surface markers and membrane integrity.
It is animal-origin-free and ready to use, with no dilution or preparation required, and it avoids the adventitious agent risk that comes with animal-derived enzymes.
Recommended use: Routine passaging and single-cell suspension preparation for primary cells, mesenchymal stem cells, iPSCs, and other adherent cell lines.
Applications
- Routine passaging
- Single-cell suspensions for counting
- Flow cytometry and cell sorting
- Cell surface marker analysis
- Viral transfection assays
- Bioreactor scale-up
- Dissociation of cells from primary tissue
- Proliferation and apoptosis assays

Protocol
Incubation time depends on cell type and density. Watch the cells, not the clock.
- 1Pre-warm. Bring the solution to room temperature or 37°C.
- 2Remove medium. Aspirate the growth medium from the culture vessel.
- 3Rinse. Rinse the monolayer briefly with HBSS without calcium or magnesium (KM-HBSS-1), DPBS (KM-DPBS-1), or serum-free medium.
- 4Add. Add enough solution to cover the cell layer, typically 2 to 5 mL per T-75 flask.
- 5Incubate. Incubate at 37°C for 3 to 10 minutes, checking detachment under the microscope.
- 6Tap. Tap the vessel gently to release remaining cells.
- 7Neutralize. Neutralize with Universal Neutralization Buffer (KM-UNB-1).
- 8Collect. Centrifuge at 200 to 300 × g for 5 minutes.
- 9Resuspend. Resuspend in fresh culture medium at the desired density.
Quality Control
Every lot is tested before release:
- Sterility: no growth after 14 days
- Mycoplasma: negative
- Endotoxin: ≤ 0.5 EU/mL
- pH and osmolality verified against specification
- Functional assay: pass (validated on multiple cell lines)
Lot-specific results are on the Certificate of Analysis. Email us to request the CoA and include your lot number from the bottle or vial label.

Frequently Asked Questions
How long is it good at 2°C to 8°C?
Up to 2 months. For longer storage, aliquot into single-use volumes and keep at −20°C.
My package was warm when it arrived. Is it still usable?
Brief ambient exposure of 24 to 48 hours is acceptable. If it was out longer than that, contact us before use.
Can I freeze it?
Yes, at −20°C for long-term storage. Aliquot into single-use volumes first so you avoid repeated freeze-thaw cycles.
How much do I need per flask?
Enough to cover the cell layer. For a T-75, that is typically 2 to 5 mL.
Does it work on iPSCs and MSCs?
Yes. It is suitable for primary cells, MSCs, iPSCs, and other adherent lines. Incubation time varies by cell type, so monitor detachment under the microscope.
