Upon Receipt
Frozen Vial: Transfer immediately to liquid nitrogen for long-term storage. 
Flask: Incubate at 37°C for 1 hour, then replace with fresh medium.  Allow cells to recover for 24 hours before sub-culturing.

The Conditionally Immortalized Human Adipose-Derived Mesenchymal Stem Cell (HAdM1) line was generated by lentiviral infection of primary HAdMSCs with an SV40 Large T antigen under a Tet-On inducible expression system. Cells were selected using puromycin resistance to ensure stable integration. The resulting line retains key MSC characteristics while providing extended proliferative capacity for research applications.

Cells are isolated from lipoaspirate derived from white adipose tissue and display a characteristic spindle-shaped, fibroblast-like morphology. This line is suitable for mechanistic studies of MSC biology, adipogenic and osteogenic differentiation research, and applications where consistent, long-term cell availability is required.

Kulture Scientific HAdM1 cells are quality-tested prior to release for surface marker expression, viability, and freedom from mycoplasma and common viral contaminants.

Receipt Protocol

Follow the appropriate protocol based on the format in which your cells were shipped. Do not deviate from these instructions — improper handling upon receipt will compromise cell viability.

Inspect vial upon receipt. Transfer immediately to liquid nitrogen vapor phase for storage. Do not allow cells to thaw.

⚠ Do not store at −20°C or −80°C. Do not allow cells to thaw during transit. Repeated freeze-thaw cycles will compromise viability.

Inspect the flask upon receipt for integrity and visible signs of contamination (turbidity, color change, particulates).

Place flask in a 37°C, 5% CO₂ humidified incubator and equilibrate for 1 hour.

Under aseptic conditions, aspirate the transit medium and replace with fresh, pre-warmed MSCGM.

Allow cells to recover for 24 hours before subculturing, passaging, or expanding.
⚠ Do not leave cells in transit medium for extended periods. Replace medium promptly upon receipt to support cell recovery.

  • Temperature sensitivity — cells are sensitive to temperature fluctuations; minimize time outside cryogenic storage
  • Freeze-thaw cycles — repeated freeze-thaw cycles will compromise cell viability and are not recommended; thaw only what is needed for immediate use
  • Stability — cells stored in liquid nitrogen are stable indefinitely when maintained under correct cryogenic conditions
  • Shipping condition — frozen vials are shipped on dry ice; T-25 flasks are shipped at ambient temperature and should be processed immediately upon receipt
  • Certificate of Analysis — lot-specific QC results including viability, sterility, and characterization data are available upon request

Biosafety Level

This cell line should be handled under Biosafety Level 2 (BSL-2) conditions. BSL-2 is required based on the following characteristics of this product:

  • Human-derived origin — all human-derived cell lines require BSL-2 containment under Universal Precautions regardless of donor screening status
  • Lentiviral transduction history — the line was generated using lentiviral vectors; replication-incompetent lentivirally transduced lines are classified as BSL-2 by most institutional biosafety committees
  • SV40 Large T antigen expression — expression of an oncogenic element requires appropriate containment

Users must consult their institutional biosafety committee (IBC) prior to use. Individual institutions may impose additional requirements based on local policy.

Biosafety Considerations
  • Human-derived material — treat as potentially infectious; follow your institutional BSL-2 guidelines for handling human-derived primary cells and cell lines
  • Lentiviral transduction — this cell line was generated using lentiviral infection; handle in accordance with your institutional biosafety committee (IBC) guidelines for lentivirally transduced cell lines
  • DMSO (cryoprotectant) — cryopreserved vials contain DMSO, which is a skin-penetrating agent; wear nitrile gloves and avoid contact with skin during thawing procedures
  • Disposal — decontaminate all cell culture waste with an appropriate disinfectant (e.g., 10% bleach) prior to disposal in accordance with local biohazardous waste regulations
Frequently Asked Questions
What makes the HAdM1 line "conditionally immortalized"?
HAdM1 cells express SV40 Large T antigen under a Tet-On inducible system. This allows proliferative capacity to be conditionally regulated by the presence or absence of doxycycline, giving researchers control over the immortalization state. Without induction, cells may revert toward more primary-like behavior, providing experimental flexibility not available with constitutively immortalized lines.
What culture medium should I use?
The recommended medium is Mesenchymal Stem Cell Growth Medium (MSCGM). Pre-warm medium to 37°C before adding to cells. Kulture Scientific offers compatible MSCGM and supplementary reagents — see the Catalog tab for catalogue numbers. Do not use DMEM alone without appropriate MSC-specific supplements, as nutrient requirements differ from standard fibroblast lines.
How long should I wait before subculturing after receipt?
Allow at least 24 hours for cell recovery regardless of format received — whether thawed from a frozen vial or transferred from a T-25 flask. Cells require time to re-adhere, equilibrate to culture conditions, and recover from transit stress before passaging. Subculturing too early will result in lower viability and reduced expansion efficiency.
Can I store cells at −20°C?
No. Neither −20°C nor −80°C is a suitable storage temperature for cryopreserved cells. Ice crystal formation at these temperatures causes irreversible membrane damage and loss of viability. Cells must be stored in liquid nitrogen vapor phase (−150°C to −196°C), where they are stable indefinitely.
Where can I find the Certificate of Analysis for my lot?
Certificates of Analysis are available upon request. Contact Kulture Scientific with the lot number printed on your vial or flask label to receive the corresponding QC documentation, including flow cytometry data, mycoplasma testing, viral contaminant screening, and viability results.
Is this product suitable for clinical or therapeutic use?
No. HAdM1 cells are for research use only and are not validated for diagnostic or therapeutic procedures. This product has not undergone the regulatory review, manufacturing standards, or safety testing required for clinical-grade cell products. Do not use in human subjects.
