KCL-ENG-103 is a stable, clonal HEK293 cell line engineered to express full-length human CD19 (Gene ID: 930) via lentiviral transduction. Following puromycin selection and limiting dilution cloning, a high-expressing clone was isolated for consistent, uniform CD19 surface expression across passages.

CD19 is expressed as a full-length surface glycoprotein (95 kDa) without fusion tags, preserving native conformation for antibody recognition and functional assay work. Greater than 90% of viable cells express human CD19 by flow cytometry, with expression stable for a minimum of 15 passages under continuous puromycin selection.

This cell line is well-suited for CAR-T cell cytotoxicity assays, ADCC studies, antibody screening, and flow cytometry standards. All lots are quality-tested prior to release for CD19 expression, post-thaw viability, mycoplasma, and sterility.

Recommended Medium

DMEM supplemented with 10% heat-inactivated FBS and 1% Pen/Strep. For routine maintenance under selection, add Puromycin at 1.0 μg/mL. Allow cells to recover in selection-free medium for 48 hours after thawing before reintroducing Puromycin. During co-culture assays with immune cells, omit Puromycin — it is toxic to lymphocytes.

Inspect the vial upon receipt. Confirm dry ice is present and the vial cap is intact. Do not allow cells to thaw.

Transfer immediately to liquid nitrogen vapor phase (−150°C to −196°C) for long-term storage.
⚠ Do not allow cells to thaw during transit or before intended use. Do not store at −80°C or −20°C.
⚗  Thawing & recovery

Thaw vial rapidly in a 37°C water bath for 60 to 90 seconds with gentle agitation. Remove when a small ice crystal remains.

Transfer cells dropwise into 10 mL of pre-warmed DMEM + 10% FBS (no Puromycin). Centrifuge at 200 × g for 5 minutes.

Aspirate supernatant and resuspend in complete medium + 0.5 μg/mL Puromycin. Seed into a T-25 flask at 37°C, 5% CO₂.

After 48 hours, increase Puromycin to 1.0 μg/mL for routine maintenance.
⚠ Do not add Puromycin at full concentration immediately after thaw. A 48-hour low-selection recovery window is required. During co-culture assays with immune cells, omit Puromycin entirely — it is toxic to lymphocytes.
Subculture Guidelines

Subculture at 80 to 90% confluence using 0.25% Trypsin-EDTA. Split 1:3 to 1:5 every 2 to 3 days. Doubling time is approximately 24 hours. Maintain under Puromycin selection (1.0 μg/mL) to preserve stable CD19 expression. Full protocols are available on the Kulture Scientific protocol page.

  • Storage — store in liquid nitrogen vapor phase (−150°C to −196°C); this is the only recommended storage condition for cryopreserved cells
  • Do not store at −80°C or −20°C — neither temperature is suitable for long-term cell storage; ice crystal formation at these temperatures will irreversibly damage cell membranes and compromise viability
  • Temperature sensitivity — minimize time outside cryogenic storage; cells are sensitive to temperature fluctuations
  • Freeze-thaw cycles — repeated freeze-thaw cycles compromise cell viability and CD19 expression stability; thaw only the quantity needed for immediate use
  • Stability — cells stored in liquid nitrogen are stable indefinitely when maintained under correct cryogenic conditions
  • Shipping condition — cryopreserved vials are shipped on dry ice; inspect packaging upon receipt and transfer to liquid nitrogen storage immediately

Biosafety Level

Frequently Asked Questions
Can I use this cell line in co-culture assays with T cells or NK cells?
Yes — KCL-ENG-103 is specifically designed for co-culture assays including CAR-T cytotoxicity and ADCC. However, Puromycin must be omitted from the co-culture medium, as it is toxic to lymphocytes. Verify CD19 expression periodically in extended experiments, particularly if cells are maintained without selection prior to the assay.
When should I add Puromycin after thawing?
Allow 48 hours of recovery in selection-free medium after thawing before adding Puromycin. Start at 0.5 μg/mL for the first 48 hours post-thaw, then increase to the maintenance concentration of 1.0 μg/mL. Adding selection antibiotic immediately after thaw causes unnecessary stress and reduces expansion efficiency.
How do I confirm CD19 expression in my lab?
Expression is confirmed by flow cytometry using an anti-human CD19-PE antibody (clone HIB19 or equivalent). Greater than 90% of viable cells should be positive, with MFI greater than 10-fold over the isotype control. Run parental HEK293 cells as a negative control. Western blot detecting the 95 kDa band is also suitable for confirmation.
How long is CD19 expression stable?
Expression has been validated as stable for a minimum of 15 passages under continuous Puromycin selection (1.0 μg/mL). If cells are maintained without selection, transgene silencing or loss may occur over time. Always maintain under Puromycin pressure and verify expression periodically if extended culture is planned.
Can I store cells at −80°C?
No. Neither −80°C nor −20°C is suitable for long-term cell storage. Ice crystal formation at these temperatures will irreversibly damage cell membranes and compromise viability. Cells must be stored in liquid nitrogen vapor phase (−150°C to −196°C), where they are stable indefinitely.
